Sample Preparation
We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. Principle of the Assay: BMP15 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-BMP15 antibody and an BMP15-HRP conjugate. The assay sample and buffer are incubated together with BMP15-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the BMP15 concentration since BMP15 from samples and BMP15-HRP conjugate compete for the anti-BMP15 antibody . Since the number of sites is limited, as more sites are occupied by BMP15 from the sample, fewer sites are left to bind BMP15-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The BMP15 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a member of the bone morphogenetic protein family which is part of the transforming growth factor-beta superfamily. The transforming growth factor-beta superfamily includes large families of growth and differentiation factors. It is thought that this protein may be involved in oocyte maturation and follicular development as a homodimer or by forming heterodimers with a related protein, Gdf9. Defects in this gene are the cause of ovarian dysgenesis 2.[provided by RefSeq, Sep 2009]

Defects in BMP15 are the cause of ovarian dysgenesis type 2 (ODG2); also known as X-linked hypergonadotropic ovarian dysgenesis or hypergonadotropic ovarian failure due to ovarian dysgenesis. Ovarian dysgenesis leads to ovarian failure and accounts for about half of the cases of primary amenorrhea. Defects in BMP15 are the cause of premature ovarian failure type 4 (POF4). An ovarian disorder defined as the cessation of ovarian function under the age of 40 years.
