The ELISA analytical biochemical technique of the MBS167553 kit is based on OPRK1 antibody-OPRK1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect OPRK1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, OPRK1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This sandwich kit is for the accurate quantitative detection of human Kappa-type Opioid Receptor (also known as OPRK1) in serum, plasma, cell culture supernates, cell lysates, tissue homogenates.

Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with human OPRK1 antibody. OPRK1 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated human OPRK1 Antibody is added and binds to OPRK1 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated OPRK1 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of human OPRK1. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

This gene encodes an opioid receptor, which is a member of the 7 transmembrane-spanning G protein-coupled receptor family. It functions as a receptor for endogenous ligands, as well as a receptor for various synthetic opioids. Ligand binding results in inhibition of adenylate cyclase activity and neurotransmitter release. This opioid receptor plays a role in the perception of pain and mediating the hypolocomotor, analgesic and aversive actions of synthetic opioids. Variations in this gene have also been associated with alcohol dependence and opiate addiction. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. A recent study provided evidence for translational readthrough in this gene, and expression of an additional C-terminally extended isoform via the use of an alternative in-frame translation termination codon.
