The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Laminin Alpha 3 (LAMa3). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Laminin Alpha 3 (LAMa3). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Laminin Alpha 3 (LAMa3), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Laminin Alpha 3 (LAMa3) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

The protein encoded by this gene belongs to the laminin family of secreted molecules. Laminins are heterotrimeric molecules that consist of alpha, beta, and gamma subunits that assemble through a coiled-coil domain. Laminins are essential for formation and function of the basement membrane and have additional functions in regulating cell migration and mechanical signal transduction. This gene encodes an alpha subunit and is responsive to several epithelial-mesenchymal regulators including keratinocyte growth factor, epidermal growth factor and insulin-like growth factor. Mutations in this gene have been identified as the cause of Herlitz type junctional epidermolysis bullosa and laryngoonychocutaneous syndrome. In nine patients with severe generalized JEB, detection of two mutations in one of the genes LAMA3, LAMB3, or LAMC2 was not possible, so the molecular basis of disease could not be clarified completely
