Reacts with the ~60kD human BCL-3 protein. Faint, non-specific bands may also be observed at ~30 and 36kD. Reactivity has been confirmed in Western blotting with REH human acute lymphocytic leukemia (ALL)(non-T, non-B), Nalm-6 human Pre-B peripheral blood leukemia, and Jurkat T lymphocyte leukemia lysates. This antibody also immunoprecipitated the BCL-3 protein from REH human acute lymphocytic leukemia lysates.

Supplied as a liquid in PBS, pH 7.4, 0.1% sodium azide, before the addition of 40% glycerol.

May be stored at 4 degree C for short-term only. For long-term storage, aliquot and store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Small volumes of anti-BCL3 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

BCL-3, a putative oncoprotein and member of the IkB family of NF-kB inhibitors, is upregulated transcriptionally in some cases of human B-cell chronic lymphocytic leukemia due to its location next to the breakpoint junction of a t(14;19) translocation(1). BCL-3 has properties of a transcriptional coactivator, bridging transcription factors with the basal transcription machinery. BCL-3 associates with the general transcription factors TFIIB, TATA-binding protein (TBP), TFIIA, CBP/p300, SRC-1, AP-1, and RXR, potentiating their activites(2-3). BCL-3 expression has been correlated with increased cellular proliferation and survival. However, in T cells, BCL-3 expression does not alter cell growth, but instead promotes cell survival. BCL-3 expression in interleukin 4 (IL-4)-deprived T cells protected the cells from apoptosis. Overexpression of BCL-3 has also been shown to increase the survival rates of activated T-cells(4). In addition, the relatively high frequency of BCL-3 expression in some non-Hodgkin and Hodgkin lymphoma types raises the possibility that Bcl-3 is involved in the pathogenesis of these tumors, and may be a target of new therapies(5).

This gene is a proto-oncogene candidate. It is identified by its translocation into the immunoglobulin alpha-locus in some cases of B-cell leukemia. The protein encoded by this gene contains seven ankyrin repeats, which are most closely related to those found in I kappa B proteins. This protein functions as a transcriptional co-activator that activates through its association with NF-kappa B homodimers. The expression of this gene can be induced by NF-kappa B, which forms a part of the autoregulatory loop that controls the nuclear residence of p50 NF-kappa B. [provided by RefSeq]
