The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing ACVR2B. The ELISA analytical biochemical technique of the MBS7250486 kit is based on ACVR2B antibody-ACVR2B antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect ACVR2B antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, ACVR2B. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: ACVR2B ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-ACVR2B antibody and an ACVR2B-HRP conjugate. The assay sample and buffer are incubated together with ACVR2B-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ACVR2B concentration since ACVR2B from samples and ACVR2B-HRP conjugate compete for the anti-ACVR2B antibody . Since the number of sites is limited, as more sites are occupied by ACVR2B from the sample, fewer sites are left to bind ACVR2B-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ACVR2B concentration in each sample is interpolated from this standard curve.
