The ELISA analytical biochemical technique of the MBS8248462 kit is based on TGFBI antibody-TGFBI antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect TGFBI antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, TGFBI. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Principle of the Assay: The Mouse TGFBI ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse TGFBI in Cell Culture Supernates, Serum, Plasma. This assay employs an antibody specific for Mouse TGFBI coated on a 96-well plate. Standards and samples are pipetted into the wells and TGFBI present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse TGFBI antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of TGFBI bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background: Transforming growth factor, beta-induced, 68kDa, also known as TGFBI (initially called BIGH3, BIG-H3), is a protein which in humans is encoded by the TGFBI gene. It is mapped to 5q31.1. This gene encodes an RGD-containing protein that binds to type I, II and IV collagens. The RGD motif is found in many extracellular matrix proteins modulating cell adhesion and serves as a ligand recognition sequence for several integrins. This protein plays a role in cell-collagen interactions and may be involved in endochondrial bone formation in cartilage. The protein is induced by transforming growth factor-beta and acts to inhibit cell adhesion.
