TGFBI, also named as BIGH3, is an adhesion protein which may play animportant role in cell-collagen interactions. TGFBI was first detected in a human lung adenocarcinoma cell line after stimulation by TGF-beta.TGFBI is secreted into the extracellular matrix (ECM) as an attachment protein. It functions mainly in cell adhesion, migration, proliferation,apoptosis, and angiogenesis. TGFBI expression is down-regulated in a variety of cancer cell lines and clinical tissue samples, and it can be used as a marker for breast cancer progression. Defects in TGFBI have been shown to be involved in several corneal dystrophies. Immunoblot analysis of TGFBI revealed both quantitative and qualitative differences between the genotypes. The secreted form of TGFBI migrates as a protein band of 75 kDa, corresponding to the predictedmolecularmass of the intact protein lacking only its signal peptide.

This gene encodes an RGD-containing protein that binds to type I, II and IV collagens. The RGD motif is found in many extracellular matrix proteins modulating cell adhesion and serves as a ligand recognition sequence for several integrins. This protein plays a role in cell-collagen interactions and may be involved in endochondrial bone formation in cartilage. The protein is induced by transforming growth factor-beta and acts to inhibit cell adhesion. Mutations in this gene are associated with multiple types of corneal dystrophy.
