This assay has high sensitivity and excellent specificity for detection of TGF-betaR2. No significant cross-reactivity or interference between TGF-betaR2 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between TGF-betaR2 and all the analogues, therefore, cross reaction may still exist in some cases.

We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. Principle of the assay: TGF-betaR2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-TGF-betaR2 antibody and an TGF-betaR2-HRP conjugate. The assay sample and buffer are incubated together with TGF-betaR2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the TGF-betaR2 concentration since TGF-betaR2 from samples and TGF-betaR2-HRP conjugate compete for the anti-TGF-betaR2 antibody . Since the number of sites is limited, as more sites are occupied by TGF-betaR2 from the sample, fewer sites are left to bind TGF-betaR2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TGF-betaR2 concentration in each sample is interpolated from this standard curve.

This gene encodes a member of the Ser/Thr protein kinase family and the TGFB receptor subfamily. The encoded protein is a transmembrane protein that has a protein kinase domain, forms a heterodimeric complex with another receptor protein, and binds TGF-beta. This receptor/ligand complex phosphorylates proteins, which then enter the nucleus and regulate the transcription of a subset of genes related to cell proliferation. Mutations in this gene have been associated with Marfan Syndrome, Loeys-Deitz Aortic Aneurysm Syndrome, and the development of various types of tumors.
