The ELISA analytical biochemical technique of the MBS167248 kit is based on BRAF antibody-BRAF antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect BRAF antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, BRAF. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This sandwich kit is for the accurate quantitative detection of human Serine, Threonine-protein Kinase B-raf (also known as BRAF) in serum, plasma, cell culture supernates, cell lysates, tissue homogenates.

Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with human BRAF antibody. BRAF present in the sample is added and binds to antibodies coated on the wells. And then biotinylated human BRAF Antibody is added and binds to BRAF in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated BRAF antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of human BRAF. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

This gene encodes a protein belonging to the RAF family of serine/threonine protein kinases. This protein plays a role in regulating the MAP kinase/ERK signaling pathway, which affects cell division, differentiation, and secretion. Mutations in this gene, most commonly the V600E mutation, are the most frequently identified cancer-causing mutations in melanoma, and have been identified in various other cancers as well, including non-Hodgkin lymphoma, colorectal cancer, thyroid carcinoma, non-small cell lung carcinoma, hairy cell leukemia and adenocarcinoma of lung. Mutations in this gene are also associated with cardiofaciocutaneous, Noonan, and Costello syndromes, which exhibit overlapping phenotypes.
