The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing LIG4. The ELISA analytical biochemical technique of the MBS2800038 kit is based on LIG4 antibody-LIG4 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect LIG4 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, LIG4. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. This assay employs a two-site sandwich ELISA to quantitate LIG4 in samples. An antibody specific for LIG4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any LIG4 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for LIG4 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of LIG4 bound in the initial step. The color development is stopped and the intensity of the color is measured.

The protein encoded by this gene is a DNA ligase that joins single-strand breaks in a double-stranded polydeoxynucleotide in an ATP-dependent reaction. This protein is essential for V(D)J recombination and DNA double-strand break (DSB) repair through nonhomologous end joining (NHEJ). This protein forms a complex with the X-ray repair cross complementing protein 4 (XRCC4), and further interacts with the DNA-dependent protein kinase (DNA-PK). Both XRCC4 and DNA-PK are known to be required for NHEJ. The crystal structure of the complex formed by this protein and XRCC4 has been resolved. Defects in this gene are the cause of LIG4 syndrome. Alternatively spliced transcript variants encoding the same protein have been observed. [provided by RefSeq, Jul 2008]

The LIG4-XRCC4 complex is responsible for the NHEJ ligation step, and XRCC4 enhances the joining activity of LIG4. Binding of the LIG4-XRCC4 complex to DNA ends is dependent on the assembly of the DNA-dependent protein kinase complex DNA-PK to these DNA ends.
