The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing MAN2B1. The ELISA analytical biochemical technique of the MBS2706470 kit is based on MAN2B1 antibody-MAN2B1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect MAN2B1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, MAN2B1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Mannosidase Alpha Class 2B Member 1 (MAN2B1). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Mannosidase Alpha Class 2B Member 1 (MAN2B1). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Mannosidase Alpha Class 2B Member 1 (MAN2B1), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Mannosidase Alpha Class 2B Member 1 (MAN2B1) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

This gene encodes an enzyme that hydrolyzes terminal, non-reducing alpha-D-mannose residues in alpha-D-mannosides. Its activity is necessary for the catabolism of N-linked carbohydrates released during glycoprotein turnover and it is member of family 38 of glycosyl hydrolases. The full length protein is processed in two steps. First, a 49 aa leader sequence is cleaved off and the remainder of the protein is processed into 3 peptides of 70 kDa, 42 kDa (D) and 13/15 kDa (E). Next, the 70 kDa peptide is further processed into three peptides (A, B and C). The A, B and C peptides are disulfide-linked. Defects in this gene have been associated with lysosomal alpha-mannosidosis.
