The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing ITGa2B. The ELISA analytical biochemical technique of the MBS2019592 kit is based on ITGa2B antibody-ITGa2B antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect ITGa2B antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, ITGa2B. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Integrin Alpha 2B (ITGa2B). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Integrin Alpha 2B (ITGa2B). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Integrin Alpha 2B (ITGa2B), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Integrin Alpha 2B (ITGa2B) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Alpha chain 2b undergoes post-translational cleavage to yield disulfide-linked light and heavy chains that join with beta 3 to form a fibronectin receptor expressed in platelets that plays a crucial role in coagulation. ITGA2B: Integrin alpha-IIb/beta-3 is a receptor for fibronectin, fibrinogen, plasminogen, prothrombin, thrombospondin and vitronectin. It recognizes the sequence R-G-D in a wide array of ligands. It recognizes the sequence H-H-L-G-G-G-A-K-Q-A-G-D-V in fibrinogen gamma chain. Following activation integrin alpha- IIb/beta-3 brings about platelet/platelet interaction through binding of soluble fibrinogen. This step leads to rapid platelet aggregation which physically plugs ruptured endothelial cell surface. Belongs to the integrin alpha chain family. Heterodimer of an alpha and a beta subunit. The alpha subunit is composed of an heavy and a light chain linked by a disulfide bond. Alpha-IIb associates with beta-3. Directly interacts with RNF181. 3 isoforms of the human protein are produced by alternative splicing.
