MBS281016 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the WD repeat domain phosphoinositide-interacting protein 4 (WDR45) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing WDR45. The ELISA analytical biochemical technique of the MBS281016 kit is based on WDR45 antibody-WDR45 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect WDR45 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, WDR45. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate WDR45 in samples. An antibody specific for WDR45 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyWDR45 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for WDR45 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of WDR45 bound in the initial step. The color development is stopped and the intensity of the color is measured.

WDR45: a member of the WD repeat protein family. WD repeats are minimally conserved regions of approximately 40 amino acids typically bracketed by gly-his and trp-asp (GH-WD), which may facilitate formation of heterotrimeric or multiprotein complexes. Members of this family are involved in a variety of cellular processes, including cell cycle progression, signal transduction, apoptosis, and gene regulation. This gene has a pseudogene at chromosome 4q31.3. Multiple alternatively spliced transcript variants encoding distinct isoforms have been found for this gene, but the biological validity and full-length nature of some variants have not been determined.
