The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing GnRH. The ELISA analytical biochemical technique of the MBS452908 kit is based on GnRH antibody-GnRH antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect GnRH antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, GnRH. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: The kit is a competitive inhibition enzyme immunoassay technique for the in vitro quantitative measurement of GNRH in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids.

Principle of the Assay: This assay employs the competitive inhibition enzyme immunoassay technique. A monoclonal antibody specific to GNRH has been pre-coated onto a microplate. A competitive inhibition reaction is launched between biotin labeled GNRH and unlabeled GNRH (Standards or samples) with the pre-coated antibody specific to GNRH. After incubation the unbound conjugate is washed off. Next, avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. The amount of bound HRP conjugate is reverse proportional to the concentration of GNRH in the sample. After addition of the substrate solution, the intensity of color developed is reverse proportional to the concentration of GNRH in the sample.
