The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing ADAM28. The ELISA analytical biochemical technique of the MBS914003 kit is based on ADAM28 antibody-ADAM28 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect ADAM28 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, ADAM28. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for ADAM28 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any ADAM28 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ADAM28 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ADAM28 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes a member of the ADAM (a disintegrin and metalloprotease domain) family. Members of this family are membrane-anchored proteins structurally related to snake venom disintegrins, and have been implicated in a variety of biological processes involving cell-cell and cell-matrix interactions, including fertilization, muscle development, and neurogenesis. The protein encoded by this gene is a lymphocyte-expressed ADAM protein. Alternative splicing results in two transcript variants. The shorter version encodes a secreted isoform, while the longer version encodes a transmembrane isoform. [provided by RefSeq, Jul 2008]

ADAM28: May play a role in the adhesive and proteolytic events that occur during lymphocyte emigration or may function in ectodomain shedding of lymphocyte surface target proteins, such as FASL and CD40L.
