Isocitrate dehydrogenases catalyze the oxidative decarboxylation of isocitrate to 2-oxoglutarate. These enzymes belong to two distinct subclasses, one of which utilizes NAD(+) as the electron acceptor and the other NADP(+). Five isocitrate dehydrogenases have been reported: three NAD(+)-dependent isocitrate dehydrogenases, which localize to the mitochondrial matrix, and two NADP(+)-dependent isocitrate dehydrogenases, one of which is mitochondrial and the other predominantly cytosolic. Each NADP(+)-dependent isozyme is a homodimer. The protein encoded by this gene is the NADP(+)-dependent isocitrate dehydrogenase found in the mitochondria. It plays a role in intermediary metabolism and energy production. This protein may tightly associate or interact with the pyruvate dehydrogenase complex. Immunoprecipitation (IP) of anti-IDHP antibody
IDHP was immunoprecipitated from 1mg of Jurkat cells membrane fraction, blotted with M25004 of 10 ug. Western blot was performed from the immunoprecipitate using M25004 at 1/2000 dilution. Anti-Mouse-IgG(HRP), specific to the non-reduced form of IgG, was used as secondary antibody at 1/10000 dilution. Lane 1: Jurkat cells membrane fraction. Lane2: IP product of Jurkat cells membrane fraction.
Blocking and dilution buffer and concentration:5% milk/TBST.

Flow Cytometry (FC/FACS) of anti-IDHP antibody
Flow cytometric analysis of 2% paraformaldehyde-fixed Jurkat (Human T cell leukemia cells from peripheral blood) cells labeling IDHP with M25004 at 1/200 dilution (green) compared with a mouse monoclonal IgG isotype control (black) and an unlabelled control (cells without incubation with primary antibody, blue). Goat anti-mouse IgG (FITC) at 1/300 dilution was used as the secondary antibody.
