The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing TRPV3. The ELISA analytical biochemical technique of the MBS2022919 kit is based on TRPV3 antibody-TRPV3 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect TRPV3 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, TRPV3. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Transient Receptor Potential Cation Channel Subfamily V, Member 3 (TRPV3). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Transient Receptor Potential Cation Channel Subfamily V, Member 3 (TRPV3). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Transient Receptor Potential Cation Channel Subfamily V, Member 3 (TRPV3), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Transient Receptor Potential Cation Channel Subfamily V, Member 3 (TRPV3) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

This gene product belongs to a family of nonselective cation channels that function in a variety of processes, including temperature sensation and vasoregulation. The thermosensitive members of this family are expressed in subsets of sensory neurons that terminate in the skin, and are activated at distinct physiological temperatures. This channel is activated at temperatures between 22 and 40 degrees C. This gene lies in close proximity to another family member gene on chromosome 17, and the two encoded proteins are thought to associate with each other to form heteromeric channels. TRPV3: Putative receptor-activated non-selective calcium permeant cation channel. It is activated by innocuous (warm) temperatures and shows an increased response at noxious temperatures greater than 39 degrees Celsius. Activation exhibits an outward rectification. May associate with TRPV1 and may modulate its activity. Is a negative regulator of hair growth and cycling: TRPV3-coupled signaling suppresses keratinocyte proliferation in hair follicles and induces apoptosis and premature hair follicle regression (catagen). May form a heteromeric channel with TRPV1. Interacts with TRPV1. Abundantly expressed in CNS. Widely expressed at low levels. Detected in dorsal root ganglion.
